Impacts of Dutasteride on the pharmacokinetics of other drugs: Dutasteride is a potent inhibitor of CYP3A4 and CYP3A5 isoenzymes and may increase the serum concentrations of drugs that are substrates for these enzymes, such as midazolam, cyclosporine, and terfenadine. Dutasteride does not inhibit CYP1A2, CYP2A6, CYP2B6, CYP2C9, CYP2C19, or CYP2D6 isoenzymes at clinically relevant concentrations. Dutasteride does not induce the metabolism of drugs metabolized by CYP450 enzymes.
Impacts of other drugs on the pharmacokinetics of Tamsulosin: Tamsulosin is primarily metabolized by CYP3A4 isoenzyme. The coadministration of potent CYP3A4 inhibitors (e.g., ketoconazole, itraconazole, ritonavir) with tamsulosin can increase the serum concentrations of tamsulosin and may result in hypotension. The coadministration of tamsulosin with other alpha-blockers may increase the risk of hypotension. The coadministration of tamsulosin with drugs that inhibit CYP2D6 isoenzyme (e.g., fluoxetine, paroxetine) can increase the serum concentrations of tamsulosin.
Impacts of Tamsulosin on the Pharmacokinetics of other drugs: Tamsulosin does not inhibit or induce CYP450 enzymes at clinically relevant concentrations. However, tamsulosin may interact with other drugs that lower blood pressure or affect heart rate, such as nitrates, beta-blockers, calcium channel blockers, or other alpha-blockers, and may increase the risk of hypotension or cardiac side effects.
Patients with hypersensitivity to Dutasteride, other 5-alpha reductase inhibitors, Tamsulosin (including Tamsulosin-induced angioedema), soya, peanut, or any of the other excipients, patients with a history of orthostatic hypotension, and patients with severe hepatic impairment should not take Tamsulosin-Dutasteride together.
The combination of Tamsulosin and Dutasteride is not recommended for women. There have been no research on the effects of combining Tamsulosin with Dutasteride on pregnancy, breastfeeding, or fertility. The following claims are based on data from studies involving the individual components. Fertility: In healthy men, dutasteride has been shown to impact sperm parameters (such as sperm count, volume, and motility). Reduced male fertility is a risk that cannot be ruled out. Tamsulosin Hydrochloride has not been studied for its effects on sperm counts or function.
